            Many studies demonstrated prominent roles of protein kinases in the regulation of cell differentiation, growth and development in plant system. Kinases are also implicated in different signal transduction pathways in abiotic and biotic stresses. By identifying the substrates of k...inases, wide assessment on how the intact plant cell regulates cell growth and responses to external stimuli can be disclosed. Thus, the main objectives of this study are to identify in vivo substrate of the isolated serine/threonine/tyrosine (STY) protein kinase from Arabidopsis thaliana and thus, to uncover the role of the kinase in plant system. This study involved the molecular cloning and gene expression of STY kinase in prokaryotic and eukaryotic systems. The studies of growth and plasmid stability of recombinant bacteria and yeast carrying STY kinase were also elucidated to assess growth behavior and stability of recombinant kinase in host system after prolonged culturing. To address the substrate of STY kinase, the study of protein-protein interaction via yeast two-hybrid system was performed to detect the positive interactors of the kinase and ultimately to identify the role of STY kinase in plant.

Bougainvillea, Hibiscus, Allamanda and other ornamental plants were used in this study. Effects of phloem and xylem stresses, thinning of leaves, amino oxaloacetic acid (AOA), sucrose, gibberellic acid, aluminium sulphate, cytokinin and glucose on the growth and development of landscaping plants were investigated. Flower/bract growth, flower size enlargement, longevity and vase life of ornamental flowers were evaluated. Chlorophyll content, carotinoid, fluorescence yield of bougainvillea and hibiscus sp. were found significantly difference at different treatments. Different micro-propagation (tissue culture) techniques and molecular (protein and DNA) biotechnology of bougainvillea, hibiscus and other landscape as well as ornamental plants have been discussed.

The materials were tested metal concentrations by using most advance analytical technique ICP-OES. The highest values of metals in under-ground Water were (4.08 K),(51.59 Na),(1.122 Li),(501.0 Ca),(13.15 Mg),(0.00 Be),(2.72Al),(0.049As),(0.061 Co),(0.111 Pb), ( 0.019 Cd),(0.220 Ni), (0.120 Cr) ,(0.202 Mn),(0.202 Zn),(7.110 Fe),(0.231Cu) and (0.029 Se) ugae*ml . The sodium adsorption ratio (SAR) values for all water samples were normal and within the permissible limits. The highest values of metals in soil were (215 K),(935 Na),(0.780 Li),(6019 Ca),(58.00 Mg),(0.00 Be),(0.760 Al),(0.012 As),(0.093 Co),(1.010 Pb), (0.921 Cd),(0.721 Ni), (0.230 Cr) ,(11.17 Mn),(0.990 Zn),(239.0 Fe),(3.100 Cu) and (0.093 Se) ugae*g .

            Conversion of agricultural products to bio-energy can take many forms. Among the conversion options are ethanol from cellulose and biodiesel from vegetable oils. Cellulosic ethanol can be produced from several sources including lumber waste, forest undergrowth, perennial grasses,... and stover from annual starch crops. The first paper in this dissertation focuses on the use of stover from sorghum and corn as a feedstock for cellulosic ethanol. Plant density is reviewed as a potential influence on the production of biomass. Biodiesel production can occur from multiple oil seed crops, dried distiller's grains with solubles, and yellow grease. Increasing annual oil yield from oil seed crops will contribute to the availability of biodiesel if fuel use increase does not exceed biodiesel production potential from the oil yield increase. The second chapter attempts to increase annual oil yield by testing multiple crop combinations for oil yield. Demand for oil seed crops can influence the production of them. Separation of lipids for alternative markets will leave the remaining bulk of oils for use in biodiesel production. The third chapter displays the lipid profiles of four crops.
